Abstract: 

Breast metastasis of OVARIAN CARCINOMA is rare. To date, only 39 cases have been reported in the English-language literature so far. We are herewith reporting a primary OVARIAN papillary serous cystadenoCARCINOMA metastasing to breast and epsilateral supraclavicular node in a 58 year old female as the first case in our centre, located at North-East India. The purpose of reporting this case is to emphasize the need of examining breasts in patients of OVARIAN cancer and the need to exercise meticulous care to exclude second primary at breast from possible secondaries at breast from OVARIAN cancer.

Abstract: 

Distant metastases are rare during the course of OVARIAN CARCINOMA. OVARIAN epithelial adenoCARCINOMAs rarely metastasize to the skin which is seen in less than 4% of patients. It usually presents as subcutaneous nodules and, less commonly, as inflammatory metastases which mimic cellulitis or viral or bacterial skin infections. The histology in most of the tumours is grade 1 or 2 serous papillary adenoCARCINOMAs. The prognosis in most patients is poor, thus, palliative treatment is usually indicated. We report a patient with OVARIAN CARCINOMA and widespread zosteriform skin metastases.

Abstract: 

Background: OVARIAN cancer is the second most prevalent gynecological malignancy. Its prognosis is poor with a five-year survival rate of <50% with the available therapies. There is a constant need for new biological markers. Therefore, we conducted the present study to evaluate the association between human epidermal growth factor receptor (HER2) neu and the clinicopathological features of epithelial OVARIAN cancer.Method: A prospective, observational analytic study was conducted at Fauji Foundation Hospital, Rawalpindi between November 2018 and October 2019. It was a cross-sectional study with a quantitative correlational study design.Results: We recruited 90 patients. The mean age of diagnosis was 53 ± 8.022 years; 81.1% (n = 73) had raised pretreatment CA-125 levels with stage III (56.7%, n = 51) and grade III (54.4%, n = 49) at presentation. It was seen that 24.4% (n = 22) of the tumors expressed HER2/neu, 65.6% (n = 59) were negative and 10% (n = 9) were equivocal. 72.2% had platinum sensitive disease. According to HER2/neu status, 20% HER2/neu positive patients had platinum sensitive disease and 3.3% had resistant disease; meanwhile, 47.8% HER2/neu negative patients had platinum sensitive disease and 10% were platinum resistant. HER2/neu expression was significantly associated with grade (P = 0.040) and pretreatment CA-125 levels (P = 0.032) whereas our study failed to show a significant association between stage (P = 0.383) and chemotherapy response (P = 0.055).Conclusion: The current study demonstrated that HER2/neu was positive in 24.4% of the patients, which was significantly associated with grade and pretreatment CA-125 level. However, a longer follow-up is needed for survival analysis and establishment of HER2/neu as a prognostic marker for epithelial OVARIAN cancer.

Abstract: 

Background: Our preliminary data on the protein expression of SORT1 in OVARIAN CARCINOMA tissues showed that sortilin was overexpressed in OVARIAN CARCINOMA patients and cell lines, while non-malignant ovaries expressed comparably lower amount of this protein. In spite of diverse ligands and also different putative functions of sortilin (NTR3), the function of overexpressed sortilin in OVARIAN CARCINOMA cells is an intriguing subject of inquiry. The aim of this study was, therefore, to investigate the functional role of sortilin in survival of OVARIAN CARCINOMA cell line.Methods: Expression of sortilin was knocked down using RNAi technology in the OVARIAN CARCINOMA cell line, Caov-4. Silencing of SORT1 expression was assessed using real-time qPCR and Western blot analyses. Apoptosis induction was evaluated using flow cytometry by considering annexin-V FITC binding. [3H]-thymidine incorporation assay was also used to evaluate cell proliferation capacity.Results: Real-time qPCR and Western blot analyses showed that expression of sortilin was reduced by nearly 70-80% in the siRNA transfected cells. Knocking down of sortilin expression resulted in increased apoptosis (27.5±0.48%) in siRNA-treated OVARIAN CARCINOMA cell line. Sortilin silencing led to significant inhibition of proliferation (40.1%) in siRNA-transfected Caov-4 cells as compared (p<0.05).Conclusion: As it was suspected from overexpression of sortilin in OVARIAN tumor cells, a cell survival role for sortilin can be deduced from these results. In conclusion, the potency of apoptosis induction via silencing of sortilin expression in tumor cells may introduce sortilin as a potential candidate for developing a novel targeted therapy in patients with OVARIAN CARCINOMA to mock control-transfected counterpart.

Abstract: 

Introduction: The relationship between ovulation induction and OVARIAN cancer is an important issue in gynecology.Case Presentation: The studied patient was a 32-year-old G1P1 woman with abdominal pain who was diagnosed with a large OVARIAN cyst during ovulation induction, which could possibly raise the risk of OVARIAN torsion. She had a 2-year secondary infertility and had received three cycles of ovulation induction with letrozole 6 months earlier. An emergency laparoscopy showed large complex OVARIAN cysts without torsion. We then performed a cystectomy. Based on the pathology report revealing a primary OVARIAN cystadenoCARCINOMA, the patient underwent a total abdominal hysterectomy, bilateral salpingo-oophorectomy, omentectomy, and retroperitoneal lymphadenectomy.Conclusions: The emergence of a suspected OVARIAN cyst during letrozole-induced ovulation is of great importance and needs to be investigated.

Abstract: 

Background: Cisplatin is one of the most widely used drugs in cancer chemotherapy. This drug has shown a great therapeutic advantage on the treatment of advanced OVARIAN CARCINOMA, bladder, head and neck, lung and some childhood neoplasms. Cell culture is one of the most suitable methods for the assesment of chemical’s effects on cell proliferation and growth, specially the cancer ones. This study was performed to investigate the effects of cisplatin on cancer cells growth. Materials and methods: Chinese hamster OVARIAN CARCINOMA cell line (CHO) has been grown in DMEM/F12 media supplied with 10% FCS and in the humified 5% CO2, 37°C incubator. Growth curve of these cells has been drawn in control and cells exposed to the highest dose-response followed concentration for one hour. Cisplatin cytotoxicity has been calculated using clonogenic assay. Prism© V.4 software has been used for statistical calculation and to draw related graphs on computer.Results: CHO growth curve followed the general pattern of logarithmic and plateaue phases. Doubling time of control cells was calculated equal to be 18.26±2.6 hour. Cisplatin cytotoxicity on this cell line followed the dose-response pattern up to 1 mg/ml. Cisplatin exposed concentration didn’t increase CHO cytotoxicity after then. Cisplatin exposure to CHO cells decreased the plating efficacy of these cells for 37% (p<0.05). Conclusion: Cisplatin affects CHO growth parameters at all different phases of plating efficacy, logarithmic and plateau. The most critical effect is however on the plating efficacy, which is consequently affecting other phases. This is what has been shown for the first time in this study. Such an observation might suggest cisplatin usage as an anti-implating agent for cancer cells, which may result in the inhibition of secondary tumour progression of patients. Other novel discovery of this study shows that although cisplatin cytotoxicity is following a direct dose-response pattern within the clinical concentrations of this drug, but has a limited range for such a direct relationship. Cisplatin does not have a significant cytotoxic effect in doses higher than 1 mg/ml in CHO cells. A similar comparative study in other cell lines and more deep investigation on possible mechanisms for these observations is suggested.
